通过计算和实验相结合的手段,了解荧光分子的构能关系和设计新型荧光染料
前言:
荧光分子有及其广泛的应用。从跟踪细胞内的蛋白转移,到DNA的测序。从研究细胞器的功能,到检测细胞的生与死。从标记癌症细胞让医生更清楚更干净的做切除手术,到各种光控药物和治疗手段的开发。整个生命科学的研究,非常大的程度上依赖于荧光技术。
目前各种荧光剂及其附带产品的年销售额大约100亿美元。而大多数荧光分子,都是大约一百年前的科学家们 “碰巧”合成出来的。其背后的很多机理,都亟待解决或存在争议。如何开发新一代更优异的荧光染料,更是燃眉之急。2014年诺贝尔奖得主,Stefan Hell, 也是超高分辨荧光显微技术的发明人之一,就准确的指出,荧光分子是荧光应用中瓶颈。诸多的新型应用,须要更亮,更稳定,更安全的荧光分子。
研究方向:My research is to study the fundamental structure-property relationships of organic fluorescent dyes, which can be used in a diverse range of applications, such as fluorescence imaging, solar cells, and dye lasers.
With an undergraduate education background in electronics, my personal research vision about organic dyes is to set up a complete set of molecular design rules, such that one gains the ultimate ability to molecular engineer “dial-up” optoelectronic properties to suit a given application, just like that electronic engineers can design (essentially any) integrated circuits accordingly to customers' needs.
To this end, we are using both "bottom-up" and "top-down" approaches to summarizing molecular design rules. In the "bottom-up" approach, we employ quantum chemical calculations and experimental characterizations to understand the molecular origins of a particular dye, before generalizing such knowledge to a wide range of compounds. In the "top-down" approach, we perform "data mining" in chemical databases and search patterns between molecular structures and their properties; subsequently, we validate these patterns using quantum chemical calculations, and thereby generate molecular design rules.
These rational molecular design rules enable us to develop novel fluorescent dyes with enhanced performance. We are working closely with organic chemists to make these compounds and explore their applications in-house. We are also actively looking for new collaborations to contribute our computational expertise. United, we hope to shift the chemistry of fluorophores from trial-and-error to concrete molecular engineering; we hope to bring people brighter fluorescent hopes in various application fields.

注2:新加坡国立大学还有一位同名的教授,是个大牛,做nanocrystal上转换的。本人对科研一腔热血,专注有机荧光分子,才刚刚上路。

What Your Job Will Be Like
Assistant Professor Liu Xiaogang’s group is looking for a postdoc to computationally model fluorescent organic molecules, in order to reveal their detailed working mechanism and systematically summarize molecular design rules for developing novel dyes with tailored properties.
The postdoctoral researcher will work as part of a team and interact closely with experimentalists. Research topics include (but are not limited to) developing novel fluorophores with enhanced brightness and photostability for super-resolution fluorescence imaging and two-photon fluorescence imaging, understanding the origins of molecular aggregations, and revealing detailed photophysical/chemical mechanisms of photobleaching and hydrogen bond interactions.
This job is for a one-year period, with the option for further extension.
Qualifications We Require- PhD in Physics, Chemistry, Materials Science, or a closely related field
- Published evidence of scientific contributions
- Experience with computational modeling
Qualifications We Desire- Experience with ab initio calculations, particularly in DFT and TD-DFT
- Experience with molecular dynamics
- Excellent programming skills
- Good knowledge about computational server setup and management
- Good understanding of photochemistry and photophysics
- A strong passion for research
- Good communication (both speaking and writing) skills and team spirit

Requirements:
- Must possess a good undergraduate/master degree in Chemistry/Chemical Engineering/Material Sciences/Physics
- Must possess at least one year of research experience (Honours/Masters project)
- Must be proficient in English (TOEFL above 85 or IELTS above 6)
- Good understanding of photochemistry will be strongly preferred
- Outstanding programming skills are preferred
- Basic knowledge of organic chemistry and fluorophore design will be beneficial
- A strong passion for research. I usually do not encourage students to go for PhD studies, because it is a highly challenging job. The “painstaking” PhD studies are only for those who are passionate about research and willing to devote their life to make an impact on the advancement of science and the long-term benefits of our society, and last but not least, have fun by doing so.

About Dr. Liu Xiaogang
Dr. Liu Xiaogang is currently working at Singapore-MIT Alliance for Research and Technology (SMART). He will move to Singapore University of Technology and Design as an Assistant Professor in Apr 2017. Dr. Liu received his B.Eng., M.Sc., M.Eng, and PhD from Nanyang Technological University, National University of Singapore, Massachusetts Institute of Technology (MIT) and the University of Cambridge, respectively, majored in electronics, nanotechnologies, materials sciences, and physical chemistry, respectively. As a fan of sciences and technologies, his personal research vision about fluorescent organic dyes is to set up a complete set of molecular design rules, such that one gains the ultimate ability to molecular engineer “dial-up” optoelectronic properties to suit a given application.
